In (a) the indicated ammounts of phospho-Ser 478 peptide (P) and non-phosphopeptide (N) were spotted on nitrocellulose. The antibody is phospho-specific in peptide dot blots when 10 x non-phosphopeptide is included to eliminate background binding to the unphosphorylated peptide. Thus far, only used in Western blots of recombinant, transiently expressed GFP-tagged proteins in HEK293 cells where it can recognise phosphorylation at Ser 478 in 20 μg lysate expressing GFP-PFKFB3 (b).
In (b), transiently expressed GFP-PFKFB3 was isolated from HEK293 cells treated or not with the Ser/Thr phosphatase inhibitor Calyculin A. The GFP immunoprecipitates were then dephosphorylated in vitro with lambda phosphatase where indicated in the absence and presence of EDTA as a phosphatase inhibitor then probed with pSer 478 using anti- GFP as loading control.
