CRISPR donor plasmid designed to tag USP30 (longest variants) with GFP at the C-terminus by an in-frame insertion upstream of the stop codon in exon 13. To be used in co-transfected with 2 guide RNAs DU48612 and DU48521 and should function with either wild-type or D10A nickase Cas9. Sequence of insert shows the donor including flanking homology arms, several silent mutations are present to prevent recognition and attack of the donor by Cas9 during the recombination procedure.
