HEK293T cells were co-transfected for 48 hours with FLAG-IRAK1 and HA-IRF5 wildtype and HA-IRF5 S362A. 20 ug of cell lysate protein were subjected to SDS-Page and immunoblotted overnight with 1 ug/ml of anti-IRF5 phospho Ser 462 plus 20 ug/ml of the unphosphorylated form of the peptide immunogen or for one hour with anti-HA (Sigma-Aldrich).
