Cells transfected with HA-TBC1D1 were stimulated with IGF1, EGF and PMA in the presence or absence of various kinase inhibitors. HA-TBC1D1 was immunoprecipitated from cellular extracts using anti-HA and subjected to SDS-Page followed by immunoblotting with anti-HA, anti-TBC1D1 phospho Ser 237 [S131C] and anti- TBC1D1 phospho Thr 596 [S112C] antibodies. Binding of the primary antibody was detected using rabbit peroxidase conjugated anti- sheep IgG antibody (1 in 10, 000 dilution, Pierce) followed by enhanced chemiluminescence (ECL, Amersham).
