Reporter construct based on lentiviral expression plasmid pLenti6/V5-D-TOPO (Invitrogen) containing Renilla luciferase under control of CMV promoter and the synthetic firefly luciferase (hlucP+) downstream of mcs for promoter sequence inserts.
Rluc cloned in by TOPO cloning downstream of CMV promoter.
MCS cloned in at unique Age1 site within V5 epitope sequence. (Age1 site at Sal1 end of MCS destroyed so Age1 between EcoR1 and Mlu1 sites available for use).
hlucP+ (with internal EcoR1 site killed) cloned in at unique SacII site.
MCS for promoter inserts : Sal1-Pac1-Nhe1-EcoR1-Age1-Mlu1
